Στα πλαίσια της παρούσας εργασίας μελετήθηκε η επίδραση της συν-έκφρασης σε κύτταρα
Escherichia coli BL21 (DE3) των μοριακών συνοδών (GroES, DnaK, Trigger factor) και της
καρβονικής αφυδατάσης (CA) φυτικής, ή ζωικής ή βακτηριακής προέλευσης, στα επίπεδα
διαλυτού κλάσματος των τελευταίων. Επιπρόσθετα, μελετήθηκε η συν-έκφραση των
μοριακών συνοδών και της χαρπίνης (HrpZ) στα επίπεδα διαλυτού κλάσματος της
ανασυνδυασμένης πρωτεΐνης.
Τα αποτελέσματα έδειξαν μια σημαντική αύξηση του διαλυτού κλάσματος της πρωτεΐνης
HrpZ, γνωστή ως χαρπίνη, με την συνέκφραση των μοριακών συνοδών, ιδιαίτερα με τις
πρωτεΐνες GroEL/GroES και με τις GroEL/GroES-DnaK/DnaJ/GrpE (συνδυασμένη δράση).
Η συν-έκφραση της καρβονικής αφυδατάσης ζωικής προέλευσης (CAC.elegans) και των
μοριακών συνοδών δεν είχε ως αποτέλεσμα σημαντική αύξηση του διαλυτού κλάσματος της
πρωτεΐνης. Η συν-έκφραση της φυτικής καρβονικής αφυδατάσης (CAArabidopsis) και των
μοριακών συνοδών GroEL/GroES και της πρωτεΐνης Trigger factor συνέβαλε στην αύξηση
του διαλυτού κλάσματος της CAArabidopsis. Η συν-έκφραση της βακτηριακής καρβονικής
αφυδατάσης (CAAzotobacter) και των μοριακών συνοδών GroEL/GroES-DnaK/DnaJ/GrpE αλλά
και των GroEL/GroES-Trigger factor συνέβαλε στην αύξηση του διαλυτού κλάσματός της.
Ακολούθησε η απομόνωση της βακτηριακής καρβονικής αφυδατάσης με τη μέθοδο της
χρωματογραφίας συγγενείας και η ανάλυση σε αποδιατακτική πηκτή ακρυλαμίδης πριν και
μετά τη συνέκφραση των μοριακών συνοδών. Επίσης, διερευνήθηκε η πιθανή αλληλεπίδραση
της καρβονικής αφυδατάσης, βακτηριακής προέλευσης, με τις πρωτεΐνες-μοριακές συνοδούς
GroEL/GroES μέσω της διαδικασίας διασύνδεσης των πρωτεϊνών με φορμαλδεΰδη.
Escherichia coli has been successfully used as a workhorse for high level production of
numerous proteins. However, overproduction of heterologous proteins in E.coli often results
in the misfolding of the protein of interest and its subsequent degradation by cellular
proteases or its deposition into biologically inactive aggregates known as inclusion bodies.
There are various approaches to minimize inclusion body formation such as co-expression of
molecular proteins or foldases (the Trigger factor, the DnaK-DnaJ-GrpE and GroEL/GroES
systems) which can be effective by increasing the soluble expression of heterologous proteins.
The aim of this study is to examine the effects of coexpression of molecular chaperones on
the production of three carbonic anhydrases from Azotobacter vinelandii (bacterial),
Arabidopsis thaliana (plant) and Caenorhabditis elegans (animal carbonic anhydrase) and
one harpin from Pseudomonas syringae pv. Phaseolicola (bacterial), in order to develop a
highly productive recombinant E.coli strain.
The process required transformation of E.coli BL21 DE3 cells with the TaKaRa plasmids
pGro7, pGkJE8, pGTf2, pKJE7 pTf16 and the plasmids containing the structural carbonic
anhydrase and harpin genes. Moreover, we examined the level of interaction of CA with the
molecular chaperones through the process of crosslinking. It was found that the soluble
fraction of recombinant carbonic anhydrases was slightly increased by co-expression with the
molecular chaperones, whereas the co-expression of molecular chaperones with harpin
resulted in a significant enhancement of the soluble fraction of the latter recombinant protein.
Among the TakaRa vectors examined, a better folding level was achieved by the coexpression
of DnaK/DnaJ/GrpE and GroES/GroEL proteins.
