Στην παρούσα εργασία μελετήθηκαν τα ανατομικά και ιστοχημικά
χαρακτηριστικά σπερμάτων και αρτίβλαστων του φυτού Abies cephalonica (έλατο)
με τη βοήθεια του Οπτικού μικροσκοπίου, του Ηλεκτρονικού μικροσκοπίου σάρωσης
και του μικροσκοπίου φθορισμού. Η μελέτη αυτή έδειξε ότι τα σπέρματα της
κεφαλληνιακής ελάτης παγώνουν σε θερμοκρασίες κάτω του –25 οC πράγμα που
πιθανόν οφείλεται στην ύπαρξη τερπενίων στο εσωτερικό τους. Επίσης μελετήθηκαν
οι κοτυληδόνες από ξερό και ενυδατωμένο σπέρμα και διαπιστώθηκε ότι σε εγκάρσια
τομή τους οι ενυδατωμένες έχουν μεγαλύτερους μεσοκυττάριους χώρους και
λιγότερες αποθηκευτικές ουσίες από τις ξερές. Η διάταξη των στομάτων που
βρίσκονται κυρίως στην προσαξονική επιφάνεια των κοτυληδόνων είναι άτακτη σε
σχέση με αυτή που έχουν τα στόματα στην επιφάνεια των φύλλων σε μεταγενέστερη
ηλικία. Στο εσωτερικό του ενδοσπερμίου παρατηρήθηκαν επίσης μεγάλοι
μεσοκυττάριοι χώροι που βοηθούν στην διέλευση του νερού μέσα από αυτούς, ενώ
στο εσωτερικό του σπέρματος βρέθηκαν αρκετές ποσότητες τερπενίων που παίζουν
αμυντικό κυρίως ρόλο. .εν βρέθηκαν καθόλου αλκαλοειδή και φλαβονοειδή σε
κανένα μέρος του σπέρματος ενώ ανιχνεύτηκαν μικροποσότητες αμύλου. Το
υποκοτύλιο του εμβρύου ήταν καφε-κόκκινο πράγμα που σημαίνει ότι υπάρχουν
ανθοκυάνες. Τέλος μικρή ήταν η παρουσία των λιπιδίων που βρέθηκαν μαζί με
τερπένια.
In the present study the anatomical features and the histochemistry of the seeds and
seedlings of the plant Abies cephalonica (spruce) were investigated using light and
scanning electron microscopy. This study showed that the seeds of Abies cephalonica
freeze at temperatures below -25 °C, which is probably due to the presence of large
quantities terpenes which are stored in them. We also studied the cotyledons in their
dry and hydrated form and found that in cross-section of the hydrated seeds the
intercellular spaces are larger and there is less storage of dry substances. The
arrangement of stomata, found mainly in the adaxial surface of the cotyledons, is
irregular compared to that of the stomata observed on the surface of leaves at a later
stage. Within the endosperm were also large intercellular spaces that seem to facilitate
the passage of water through them during inbibition. Inside the seed large quantities
of terpenes were found which seem to be primarily for defence. No alkaloids nor
flavonoids were found in any part of the seed and only very small amounts of starch
were found in the storage tissues. The hypocotyl of the embryo was brown-red which
means that there was a small amount of anthocyanins in the epidermal cells. Finally
lipids and terpenes were also found but in smaller amounts in the hypocotyl tissues.
