Τo MHC (Μείζον Σύστημα Ιστοσυμβατότητας) είναι σύμπλοκο γονιδίων που εντοπίζεται σε όλα τα σπονδυλωτά, με ρόλο την κωδικοποίηση μορίων που διαδραματίζουν κεντρικό ρόλο στην ανοσολογική απόκριση. Τα γονίδια του τόπου
αυτού χωρίζονται σε τρείς ομάδες (Ι, ΙΙ και ΙΙΙ) με βάση την δομή και το
λειτουργικό τους ρόλο. Ρόλος του DRB1, που ανήκει στα γονίδια τάξεως II και
είναι από τα πιο πολυμορφικά γονίδια του MHC, είναι να κωδικοποιεί
γλυκοπρωτεΐνες που προσδένονται και παρουσιάζουν στον Τ-κυτταρικό υποδοχέα
(TCR) των CD4
+
Τ-κυττάρων αντιγονικά πεπτίδια που προέρχονται κατά κύριο
λόγο από εξωκυτταρικές πρωτεΐνες και παράσιτα. Καθώς η πλειοψηφία του
νουκλεοτιδικού πολυμορφισμού της Τάξεως ΙΙ εντοπίζεται στο εξόνιο 2, μελέτες με
σκοπό την συσχέτιση του πολυμορφισμού του MHC με ανθεκτικότητα σε
παρασιτώσεις εστιάζονται σε αυτό.
Για τους σκοπούς του παρόντος πειράματος λήφθηκαν δείγματα αίματος και
κοπράνων από 45 Κοκοβίτικα και από 50 Αργίτικα πρόβατα. Απομονώθηκε
γονιδιωματικό DNA από κάθε δείγμα αίματος και στην συνέχεια ακολούθησε
προσδιορισμός του παρασιτικού φορτίου των ζώων και της έκτασης της βλάβης του
γαστρεντερικού βλεννογόνου με μετρήσεις του αριθμού των αυγών των ενήλικων
προνυμφών των παρασίτων ανά γραμμάριο κοπράνων (FEC) και των επιπέδων του
πεψινογόνου στο πλάσμα του αίματος, αντίστοιχα. Με βάση τα ευρήματα αυτά,
επιλέχθηκαν για γονοτύπηση 12 Κοκοβίτικα πρόβατα που εμφάνιζαν τιμές FEC
από 0 έως 100 και 14 ζώα που εμφάνιζαν τιμές FEC >400. Από τη φυλή Άργους
επιλέχθηκαν 13 ζώα με τιμές FEC από 0 έως 100 και 14 ζώα με τιμές FEC>1300.
Από την αλληλούχηση ταυτοποιήθηκαν 24 αλληλόμορφα, γεγονός που υποδηλώνει
υψηλά επίπεδα πολυμορφισμού. Και στις 2 φυλές ο βαθμός ετεροζυγωτίας ήταν
πολύ υψηλός και ίσος με 0,90 για την φυλή Άργους και 0,84 για την Κοκοβίτικη
φυλή, ενώ και για τις δύο φυλές ταυτοποιήθηκαν από 17 αλληλόμορφα. Από τα 24
αλληλόμορφα που ταυτοποιήθηκαν συνολικά, τα 10 ήταν κοινά για τις 2 φυλές, ενώ
τα περισσότερα αλληλόμορφα είχαν ήδη βρεθεί σε μελέτες πολυμορφισμού του
DRB1 σε ελληνικές φυλές (Φριζάρτας, Κοζάνης, Καλαρρύτικο) συμπεριλαμβανομένων των αλληλομόρφων DRB1*1606, DRB1*2003, DRB1*1008
και DRB1*0806 που βρέθηκαν αποκλειστικά σε ελληνικές φυλές. Ανιχνεύθηκε
παρόλα αυτά 1 νέο αλληλόμορφο , το DRB1*2601, στα ζώα της φυλής Άργους. Το
νέο αλληλόμορφο παρουσιάζει ενδιαφέρον καθώς εμφανίζει καινούργιες
πολυμορφικές θέσεις, αποτελώντας έτσι μια νέα οικογένεια αλληλομόρφων. Αυτό
το αλληλόμορφο διαφέρει κατά ένα νουκλεοτίδιο με αλληλόμορφο που έχει βρεθεί
στα άγρια πρόβατα της Βορείου Αμερικής (Ovis canadensis) εγείροντας
ερωτήματα για την εξελικτική πορεία των δύο ειδών.
Για τα ζώα στα οποία βρέθηκε το νέο αλληλόμορφο DRB1 εξετάστηκε και το
γονίδιο χαμηλού πολυμορφισμού Ovar-DRA, με τα ταυτοποιηθέντα αλληλόμορφα
να είναι τα DRA*0101 και DRA*0102. Το DRA*0102 εμφανίζεται και στα άγρια
πρόβατα (Ovis canadensis), γεγονός που δεν προξενεί εντύπωση καθώς τα DRA και
DRB γονίδια είναι στενά συνδεδεμένα και κληρονομούνται μαζί.
Σχετικά με τις παρασιτολογικές παραμέτρους, τα Κοκοβίτικα πρόβατα εμφάνιζαν
χαμηλότερο παρασιτικό φορτίο από ότι τα ζώα της φυλής Άργους, καθώς τα
Κοκοβίτικα ήταν εκτρεφόμενα ημιεκτατικά σε ορεινές βοσκές με μικρότερη
πυκνότητα βοσκής, ενώ η φυλή Άργους εκτρεφόταν σε πεδινές βοσκές όπου είχαν
πρόσβαση και άλλα κοπάδια προβάτων, στα οποία δεν ακολουθείται πάντα η
σωστή αντιπαρασιτική αγωγή. Οι παρατηρούμενες τιμές πεψινογόνου δεν
σχετίζονταν με το είδος παρασίτου ούτε με τους γονότυπους ή συγκεκριμένα
αλληλόμορφα, εκτός μιας περίπτωσης όπου για ένα αλληλόμορφο, το OvarDRB1*0105,
παρατηρήθηκε αρνητική συσχέτιση με τις τιμές mI.U τυροσίνης και
είχε μεγάλη συχνότητα εμφάνισης στις ομάδες υψηλού παρασιτικού φορτίου. Οι
έλεγχοι για πιθανή συσχέτιση των διαφόρων γονοτύπων και των αλληλομόρφων με
τα παρασιτολογικά χαρακτηριστικά (χαμηλές ή υψηλές τιμές FEC) δεν έδειξαν
περαιτέρω στατιστικά σημαντικά αποτελέσματα. |

The MHC – Major Histocompatibility Complex is a genetic region within all
vertebrates that encodes molecules playing a central role in immunological
response. There are three classes of MHC genes (I, II and III) depending on their
structure and function. The role of DRB1 genes, which belong to the Class-II region,
and are amongst the most polymorphic genes in the MHC, is to encode
glycoproteins which bind and present pathogen peptide fragments to the T-cell
receptor (TRC) of circulating CD4+ T cells. These antigen peptides usually derive
from extra-cellular proteins such as parasite proteins. As the majority of nucleotide
polymorphism at Class II loci locates in the second exon, genotyping studies usually
focus on it to associate MHC diversity with resistance or susceptibility to disease.
The purpose of this study was to determine the DRB1 alleles existing in two Greek
breeds (Argos and Kokovitika) and associate them to immunological aspects, such
as nematode resistance. Genotyping Greek breeds is important, since from previous
studies, a number of new alleles was found and submitted into the Immuno-
Polymorphism Database, meaning easier future genotyping and resulting in an
updated and sufficiently diverse database. Genotyping these breeds would also help
in an effort to preserve them and maintain diversity as well as preserve potentially
rare alleles contributing to beneficial traits.
At the start of this study, blood and faecal samples were taken from 45 sheep of the
Kokovitiko breed and 50 from the Argos breed. Genomic DNA was prepared and
taken to the Moredun Institute. The DNA samples were associated with faecal egg
counts per gram (EPG) and linked gastrointestal parasite species data, quantified
and their quality was determined by nanodrop spectophotometry. The samples were
then sorted into groups of high and low EPG. 12 Kokovitika and Argos sheep with
FEC<100, as well as 14 Kokovitika with FEC>400 and 14 Argos sheep with
FEC>1300, were chosen for genotyping at the DRB1 locus by sequence based
typing. The breeding rams of the Argos breed were also genotyped.
The sequencing resulted in the identification of 24 alleles, showing high diversity
levels. For the Argos and the Kokovitika sheep, a total of 17 different alleles were
identified per breed. Ten out of these 24 alleles were common for both breeds. Most of the alleles identified were also identified previously in studies of DRB1
polymorphism in Greek breeds (Frizarta, Kozani, Boutsiko), including alleles
DRB1*1606 and DRB1*2003, DRB1*1008 and DRB1*0806 which were
specifically found in these Greek breeds.
The alleles with the highest frequency in both breeds were DRB1*1001 and
DRB1*0806. Interestingly, allele 1001 appeared in the Argos breed 11 times in the
low FEC group and only once in the high FEC group, whereas in the Kokovitika
sheep it appeared 7 times in the high FEC group but didn’t appear in the low group.
In this case, the breed has an effect on the way this allele is expressed. Allele *0105
also appeared more frequently in the high FEC group for both breeds and statistical
analysis showed it was related to higher tyrosine counts in the blood’s plasma. All
other alleles were evenly distributed within the groups and further statistical
analysis showed no statistical significance between alleles or genotypes and
parasitological parameters. Statistical analysis performed in order to relate FEC
counts with pepsinogen levels, showed no significant correlation.
The new allele found in the Argos breed samples is of interest as it presents
substantial variation and appears to have novel polymorphic sites, unrelated to all
other domestic sheep DRB1 alleles, representing a novel allelic family
(DRB1*2601). This allele appears to be closely related to alleles found in wild
bighorn sheep in North America (Ovis canadensis) raising interesting questions on
the evolution of the two sheep species.
In the Class II of the MHC of sheep (OLA) the chain of the DR heterodimer is
closely linked to DRB and considered almost monomorphic, but because allele
DRB1*2601 was very divergent, it was of interest to link it to a polymorphism in
the DRA locus, so samples were sent for bidirectional sequencing. The sequencing
reads revealed a polymorphic site but the two alleles identified were DRA*0101 and
DRA*0102. DRA*0102 also appears in wild bighorn sheep (Ovis canadensis),
which is logical since the DRA and DRB are closely linked and passed on together. |
