Στην παρούσα επιστημονική έρευνα μελετήθηκαν 20 διαφορετικά δείγματα τυριών από ποικιλίες οι οποίες παρασκευάστηκαν στην περιφέρεια «Basilicata» της Νότιας Ιταλίας, και ανήκουν στους τύπους ‘pecorino’ (Canestrato di Moliterno, Pecorino Abriola, Filiano, Lucano) και ‘pasta-filata’(Caciocavallo Lucano, Caciocavallo Podolico, Caciocavallo Silano, Provolone Lucano, Provolone semipicante, Scamorza), διαφορετικών παραγωγών και χρόνων ωρίμανσης. Η μικροβιακή ποικιλότητα των στελεχών που απομονώθηκαν και η τυπική φυσικοχημική σύσταση προσδιορίστηκαν με τη χρήση προτύπων αναλυτικών τεχνικών, με συνδυασμό κλασσικών μεθόδων και ανάλυση αλληλουχίας. Χρησιμοποιήθηκε επίσης τροποποιημένο mMRS agar το οποίο περιείχε Κυανούν της Βρωμοφαινόλης και Κυστεΐνη για τη διαφορική καταμέτρηση των οξυγαλακτικών βακτηρίων σε μικτή καλλιέργεια, και να οδηγήσει τη μοριακή ταυτοποίηση. Καταμετρήθηκε επίσης ο NSLAB πληθυσμός των μεσόφιλων γαλακτοβακίλλων, όπως και ο πληθυσμός των μεσόφιλων οξυγαλακτικών που πιθανώς επιβιώνει κατά τη διέλευση στο γαστρικό σωλήνα. Τέλος, σε επιλεγμένα στελέχη εξετάστηκαν ορισμένες λειτουργικές ιδιότητες όπως η αντοχή σε χολικά άλατα, η ικανότητα συσσωμάτωσης και η δραστηριότητα υδρόλυσης των χολικών αλάτων.
Τα τυριά pasta-filata συγκριτικά με τα pecorino έδειξαν μεγαλύτερες τιμές όπως σε όλες τις παραμέτρους που μετρήθηκαν. Οι τιμές της ενεργότητα νερού στα τυριά ‘pecorino’ κυμάνθηκαν από 0.874 έως 0.902 και στα ‘pasta-filata’ από 0.903 ως 0.956, ενώ το pH από 4.97 έως 5.25 και 4.95 με 5.6 αντίστοιχα. Ακόμη μεγάλος εύρος τιμών είχε το % υγρασίας 26.5-49.0% για τα pasta-filata εν αντιθέση του άλλου των pecorino με 28,6-32.3%, και σχεδόν αναλογικά το %αλατιού στη υδατική φάση 2.55-7.38% και 5.97-10.25% αντίστοιχα. Υψηλοί πληθυσμοί μεσόφιλων γαλακτοβακίλλων βρέθηκαν με τα δε pecorino να κυμαίνονται από 4.8-7.4 log10cfu/g και τα pasta-filata 6.2-8.6 log10cfu/g. Πιθανών να επιβιώνουν κατά τη διέλευση στο γαστρικό σωλήνα 4.8-7.4 log10cfu/g και 4.7-8.5 log10cfu/g αντίστοιχα.
Συνολικά απομονώθηκαν 325 αποικίες, εκ των οποίων οι 27 (8.3%) ήταν cat(+) και 14 (4.3%) δεν κατάφεραν να αναπτυχθούν σε MRS broth pH 6.8. Εφαρμόστηκε η τεχνική RAPD-PCR σε 284 στελέχη με τους εκκινητές Μ13 για γαλακτοβακίλλους (78.8%) και coc-1 για τους κόκκους (18.3%). 8 (2.9%) από τα στελέχη δεν ενισχύθηκαν. Με τη χρήση UPGMA κατάφεραν να διαχωριστούν τα στελέχη, ενώ καλύτερη ταξινόμηση παρουσιάστηκε μόνο για τους γαλακτοβάκιλλους. Λήφθησαν 162 διαφορετικά RAPD-προφίλ, και τα μοναδικά στελέχη αυτά ταυτοποιήθηκαν με multiplex-PCR για το Lactobacillus casei group, και Lb. plantarum group, ITS-PCR, με ενίσχυση και αλληλούχιση του γονιδίου του 16S rRNA όπως και με συνδυασμό φαινοτυπικών και γενοτυπικών μεθόδων.
Η ταυτοποίηση των ειδών έδειξε ότι ο οξυγαλακτικός πληθυσμός αποτελείται κατά 48.5% (134 στελέχη) από βακτήρια του Lactobacillus casei group (Lb. paracasei/rhamnosus). Τα άλλα ταυτοποιηθέντα ανήκουν σε άλλα είδη γαλακτοβάκιλλων και κόκκων. Όσον αφορά τυριά τα pasta-filata, τα περισσότερα στελέχη ταυτοποιήθηκαν επιτυχώς σαν Lactobacillus paracasei και Lb. rhamnosus. Συγκεκριμένα 11 στελέχη από τα 216 ταυτοποιήθηκαν ως Enterococcus faecium (5.1%), 4 ως Lactobacillus buchneri (1.85), 1 ως Lb. curvatus (0.5%), 13 ως Lb. delbrueckii subsp delbrueckii (6.0%), 48 ως Lb. fermentum (22.22%), 13 ως Lb. parabuchneri (6.0%), 78 ως Lb. paracasei (36.1%), 30 ως Lb. rhamnosus (13.9%), 2 ως Lb. spp (Lspp), 3 ως Lactococcus lactis (Lcl), 1 ως Lb. casei (Lca), 1 ως Lb. plantarum (0.5%), 4 ως Lb. rhamnosus/paracasei (1.8%), 5 ως Pediococcus acidilactici (2.3%), 1 ως P. pentosaceus (0.5%), και 1 ως Streptococcus thermophillus (0.5%). Όσον αφορά τη σύσταση των ‘pecorino’ τυριών (από 60 ταυτοποιήσεις) 1 (1.66%) ήταν Enterococcus durans, 13 (21.66%) E. faecium, 1 (1.66%) E. faecium/faecalis, 1 (1.66%) E. mundtii, 1 (1.66%) Lactobacillus brevis, 19 (31.66%) Lb. paracasei, 6 (10%) Lb. rhamnosus, 11 (18.33%) Pediococcus acidilactici, 6 (10%) P. pentosaceus, και 1 (1.66%) Streptococcus macedonicus.
Η αξιολόγηση πιθανών λειτουργικών ιδιοτήτων (δυνητικής προβιοτικής δράσης) έγινε σε ταυτοποιημένα στελέχη των ειδών Lb. paracasei, Lb. rhamnosus, και Lb. plantarum. Η πλειοψηφία των στελεχών (64%) καθέστη ανίκανη να αναπτυχθεί και να σχηματίσει αποικίες ακόμη και στην ελάχιστη συγκέντρωση των χολικών αλάτων (0.3%). Tα ποσοστά ανθεκτικότητας των στελεχών Lb. rhamnosus και Lb. paracasei ήταν σχεδόν ίδια. Μόλις εφτά από τα στελέχη κατάφεραν να αναπτυχθούν κάτω από όλες τις συγκεντρώσεις χολικών αλάτων που δοκιμάστηκαν. Τα 54 (60%) δεν έδειξαν συσσωμάτωση (aggregation) είτε στον σωλήνα MRS broth pH 6.8, είτε κάτω από μικροσκοπική παρατήρηση, ενώ 25 (27.8%) έδειξαν θετική δοκιμή κάτω και από τα δυο τεστ. Το ποσοστό των θετικών στελεχών στην δοκιμή της υδρόλυσης των χολικών αλάτων είναι σχεδόν ίδια για τα δυο πιο πολυάριθμα στελέχη που ελέγχθηκαν (Lb. paracasei και Lb. rhamnosus) με περίπου το 39% των στελεχών θετικά. Τέλος το στέλεχος Lb. plantarum ήταν το πιο ανθεκτικό κατά τον έλεγχο αντοχής σε χολικά άλατα σε όλες τις συγκεντρώσεις, ενώ παρουσίασε και δραστηριότητα-BSH, αλλά δεν έδειξε ικανότητα συσσωμάτωσης.
Εν κατακλείδι το διαφορικό υπόστρωμα mMRS-BPB αποτέλεσε ένα χρήσιμο εργαλείο οδήγησης της μοριακής ταυτοποίησης με μοριακές μεθόδους βασισμένες στην PCR, χωρίς να είναι εύκολη η διαφορική καταμέτρηση σε μίγμα αποικιών. Επίσης δεν καθέστη δυνατός ο ακριβής συνδυασμός συγκεκριμένης αποικίας με συγκεκριμένο είδος.
In the present research, 20 different cheese samples from different varieties and
periods of ripening, which produced in the Region of Basilicata (Southern Italy),
collected from different producers. They belong to the big families of ‘pecorino’
(Canestrato di Moliterno, Pecorino Abriola, Filiano, Lucano) and ‘pastafilata’(Caciocavallo
Lucano, Caciocavallo Podolico, Caciocavallo Silano, Cacio
Bucato, Provolone Lucano, Provolone semipicante, Scamorza). The microbial
diversity of the isolated strains and the typical physicochemical composition
determined utilizing standard analytical techniques, with the combination of classical
methods and sequence-analysis. A modified mMRS agar, which contained
Bromophenol blue and Cysteine was used for the differential measurement of the
lactic acid bacterium in mixed culture, and therefore to lead to the molecular
identification. Furthermore, it was measured the population of mesophilic NSLAB
lactobacilli and the population of the mesophilic lactic acid bacteria (LAB), which
were probably able to survive under treatment with simulated gastric juice. Finally, in
selected strains some functional properties have been examined, such as the resistance
in bile salts, the aggregation ability and the BSH-activity (Bile Salt Hydrolysis).
The pasta-filata cheeses, compared to pecorino cheese, have shown higher values in
all the measured parameters. The water activity values for 'pecorino' cheeses ranged
from 0.874 to 0.902 and for 'pasta-filata' from 0.903 to 0.956, while the pH from 4.97
to 5.25 and from 4.95 to 5.6 respectively. Also, a wide range of values noticed for the
percentage of moisture, with 26.5-49.0% for 'pasta-filata' unlike to 28.6-32.3% for
'pecorino' and almost proportional the percentage of salt in moisture (s/m) with 2.557.38%
and 5.97-10.25% respectively. In addition, high populations of mesophilic
NSLAB lactobacilli found in both types ranging from 4.8-7.4 log10cfu/g in pecorino
cheeses and in pasta-filata 6.2 to 8.6 log10cfu/g. Almost equal numbers had the
mesophilic LAB, which were able to survive after the treatment with simulated gastric
juice both in pecorino and pasta-filata, with the first reaching numbers from 4.8 to 7.4
log

cfu/g and the second from 4.7 to 8.5 log
cfu/g.
Totally, 325 colonies with different morphologies were isolates randomly from the
last plate dilution. However, 27 (8.3%) were cat
[5]
10
(+)
and 14 (4.3%) did not manage to
survive in MRS broth pH 6.8. Randomly amplified polymorphic DNA-PCR was
applied in 284 strains with the primer M13 for lactobacilli (78.8%) and the primer
coc-1 for the cocci (18.3%). However, 8 (2.9%) show no band detection. The
“Unweighted paired-group with arithmetic averages” method (UPGMA) and
clustering algorithm was applied in order to discriminate the strains, whereas better
classification was appeared only for lactobacilli. Generally, 162 different RAPDprofiles
were obtained and these unique strains identified with species specific-PCR
for Lactobacillus casei group and Lb. plantarum group, with ITS-PCR and with
amplification and sequence of the gene of 16S rRNA region, as well as with
combination of phentotypic and genotypic methods.
The identification of the species showed the LAB microflora was consisted of
48.5% (134 strains) with bacteria of Lactobacillus casei group (Lb.
paracasei/rhamnosus). The other identified strains were other species of lactobacilli
and also some cocci.
As with pasta-filata cheeses, most of the strains successfully identified as
Lactobacillus paracasei and Lb. rhamnosus. More specifically, 11 strains from 216
identified as Enterococcus faecium (5.1%), 4 as Lactobacillus buchneri (1.85), 1 as
Lb. curvatus (0.5%), 13 as Lb. delbrueckii subsp delbrueckii (6.0%), 48 as Lb.
fermentum (22.22%), 13 as Lb. parabuchneri (6.0%), 78 as Lb. paracasei (36.1%), 30
as Lb. rhamnosus (13.9%), 2 as Lb. spp (Lspp), 3 as Lactococcus lactis (Lcl), 1 as Lb.
casei (Lca), 1 as Lb. plantarum (0.5%), 4 as Lb. rhamnosus/paracasei (1.8%), 5 as
Pediococcus acidilactici (2.3%), 1 as P. pentosaceus (0.5%), and 1 as Streptococcus
thermophillus (0.5%). Regarding the composition of ‘pecorino’ cheese (60 strains), 1
(1.66%) was Enterococcus durans, 13 (21.66%) E. faecium, 1 (1.66%) E.
faecium/faecalis, 1 (1.66%) E. mundtii, 1 (1.66%) Lactobacillus brevis, 19 (31.66%)
Lb. paracasei, 6 (10%) Lb. rhamnosus, 11 (18.33%) Pediococcus acidilactici, 6 (10%)
P. pentosaceus, and 1 (1.66%) Streptococcus macedonicus.
The assessment of potential functional properties (potential probiotic action) applied
in identified strains of the species Lb. paracasei, Lb. rhamnosus, and Lb. plantarum.
The majority of the strains (64%) were unable to grow even in the minimum
concentration of bile salts (0.3%). Also, the percentage of the Lb. rhamnosus and Lb.
paracasei strains were almost equal. Unfortunately, only seven from the strains
manage to grow under all the concentrations of biles tested. Furthermore, 54 (60%)
did not of ability of aggregation neither in MRS broth pH 6.8, nor under microscopic
examination, whereas 25 (27.8%) showed positive results under both tests. The
percentage of positive strains in the test of BSH-activity was almost the same for both
most numerous specie (Lb. paracasei and Lb. rhamnosus) that was checked, with
around 39% of the strains of each specie to be positive. Finally, the only strain Lb.
plantarum was the most resistant in all concentration of bile salts, showed remarkable
BSH-activity, but not any aggregation ability.
In conclusion the differential modified mMRS-BPB substrate was a useful tool to
drive the molecular identification with PCR-based molecular techniques, without be
easy the differential counting of colonies in mixed culture and the combination of a
specific colony with a specific specie
