Ένα από τα μεγαλύτερα προβλήματα στη βιομηχανία τροφίμων είναι η ανάπτυξη βιοϋμενίων, παθογόνων και αλλοιωγόνων βακτηρίων, τα οποία επιμολύνουν τα τρόφιμα κατά την επεξεργασία τους, καθιστώντας τα πιθανή αιτία τροφιμογενών λοιμώξεων. Η κατανόηση του τρόπου με τον οποίο σχηματίζονται και αναπτύσσονται τα βιοϋμένια θα βοηθήσει σημαντικά στον περιορισμό τους και επομένως σε σαφώς ασφαλέστερα προϊόντα. Δύο από τους πιο σημαντικούς μικροοργανισμούς που σχετίζονται με τροφιμογενείς λοιμώξεις είναι οι Staphylococcus aureus και Salmonella enterica ser. Typhimurium, και επομένως, η μελέτη της ικανότητας τους να δημιουργούν και να αναπτύσσονται σε βιοϋμένιο είναι μεγάλης σημασίας. Παράλληλα, τα τελευταία χρόνια το ενδιαφέρον έχει επικεντρωθεί και στη χρήση ωφέλιμων μικροοργανισμών με αντιμικροβιακή δραστηριότητα, με στόχο την εξασφάλιση ασφαλών προϊόντων. Ο Streptococcus macedonicus είναι ένα βακτήριο με δυνητικά μεγάλες δυνατότητες αξιοποίησης στον τομέα της βιοσυντήρησης κυρίως μέσω της παραγωγής βακτηριοσινών. Σε αυτό το πλαίσιο, αντικείμενο της παρούσας μελέτης ήταν η αξιολόγηση της ικανότητας σχηματισμού βιοϋμενίου των παθογόνων βακτηρίων Staph. aureus και S. Typhimurium, παρουσία κυττάρων ή μεταβολικών προϊόντων του βακτηρίου Str. macedonicus. Αρχικά, αξιολογήθηκε ο σχηματισμός βιοϋμενίου σε κουπόνια από ανοξείδωτο χάλυβα για τους παραπάνω μικροοργανισμούς σε μονοκαλλιέργειες, καθώς και σε μικτές καλλιέργειες του κάθε παθογόνου με τον Str. macedonicus. Η επώαση έλαβε χώρα στους 37 και 15°C για συνολικά 72 και 144 h, αντίστοιχα. Η αρχική προσκόλληση κυττάρων στα κουπόνια σε κάθε είδος καλλιέργειας αξιολογήθηκε στις 3 h επώασης στους 15°C σε αλατούχο διάλυμα, ενώ η βιοϋμενική ανάπτυξη μελετήθηκε σε θρεπτικό ζωμό κατά της διάρκεια της επώασης στις παραπάνω θερμοκρασίες. Δεν παρατηρήθηκε ιδιαίτερη επίδραση του Str. macedonicus στην ανάπτυξη των παθογόνων. Παρόλα αυτά, η ανάπτυξη του Str. macedonicus φάνηκε να ευνοείται στα μικτά βιοϋμένια, ειδικά παρουσία της S. Typhimurium, παρά το γεγονός ότι η τελευταία αρχικά παρεμπόδισε σε κάποιο βαθμό την προσκόλλησή του. Παράλληλα με την ανάπτυξη βιοϋμενίου σε κουπόνια κάτω από τις παραπάνω συνθήκες, ελήφθησαν δείγματα-κουπόνια στις 72 h για τους 37°C και στις 144 h για τους 15°C, για μέτρηση της ηλεκτρικής αγωγιμότητας σε συσκευή Malthus. Η συσκευή μετρά την ηλεκτρική αγωγιμότητα, η οποία μεταβάλλεται όσο υπάρχει ανάπτυξη και επομένως αλλάζει η συγκέντρωση των μεταβολικών προϊόντων στο διάλυμα, και δίνει την τιμή χρόνου κατά την οποία αυτή σταθεροποιείται. Δυστυχώς, το πλήθος των δειγμάτων που έδωσε μετρήσιμη τιμή χρόνου εντοπισμού ήταν χαμηλό, επομένως δεν προέκυψαν συμπεράσματα. Στη συνέχεια, μελετήθηκε η ικανότητα σχηματισμού βιοϋμενίου των παραπάνω παθογόνων σε συνθήκες μονοκαλλιέργειας παρουσία υπερκειμένου καλλιέργειας (cell-free culture supernatant, CFS) του Str. macedonicus σε συγκεντρώσεις 0, 20 και 40% (v/v), σε μικροσυστοιχίες πολυστυρενίου, και μέσω χρώσης κρυσταλλικού ιώδους και μέτρηση της οπτικής πυκνότητας (580nm). Τα αποτελέσματα φάνηκαν να συμβαδίζουν με εκείνα στην ανάπτυξη σε κουπόνια εκτός της περίπτωσης της S. Typhimurium στους 37°C και σε 40% CFS, που έδωσε μεγαλύτερη τιμή οπτικής πυκνότητας, υποδεικνύοντας μεγαλύτερη ικανότητα σχηματισμού βιοϋμενίου συγκριτικά με το μάρτυρα (0% CFS). Παρομοίως, αξιολογήθηκε η ικανότητα σχηματισμού βιοϋμενίου των παθογόνων Staph. aureus και S. Typhimurium σε συνθήκες μονοκαλλιέργειας παρουσία 0% και 40% (v/v) CFS του Str. macedonicus, σε κουπόνια ανοξείδωτου χάλυβα. Η επώαση έλαβε χώρα στους 37°C για 72 h και στους 15°C για 144 h. Η παρουσία του CFS δεν φάνηκε να επηρεάζει τη βιοϋμενική ανάπτυξη του Staph. aureus, αλλά φάνηκε να κρατά την ανάπτυξη της S. Typhimurium σε χαμηλότερα επίπεδα στο 40% από ότι στο 0% CFS. Ακόμη, στους 37οC, η προσκόλληση ήταν καλύτερη από ότι στους 15°C και η παρουσία του 40% CFS φάνηκε να επηρεάζει την προσκόλληση και των δύο παθογόνων σε σχέση με το μάρτυρα (0% CFS). Τα δεδομένα της συγκεκριμένης μελέτης υποδεικνύουν ότι τα βιοϋμένια είναι δυναμικά συστήματα, και διάφοροι παράγοντες μπορεί να επηρεάσουν σημαντικά την συμπεριφορά τους. Κατά συνέπεια, περαιτέρω έρευνα αναφορικά με την επίδραση διαφορετικών συνδυασμών ενδογενών και περιβαλλοντικών παραμέτρων στην ανάπτυξη βιοϋμενίου θα είχε μεγάλο ενδιαφέρον, και αναμένεται να συμβάλλει στην καλύτερη κατανόηση των εμπλεκόμενων στη βιοϋμενική ανάπτυξη μηχανισμών αλλά και στο σχεδιασμό αποτελεσματικών τρόπων ελέγχου.
One of the biggest problems in the food industry is biofilm formation of pathogen and spoilage bacteria, which may contaminate foods during processing, rendering them potential causes of foodborne diseases. The understanding of the way in which biofilms are being formed is going to help significantly in their limitation and therefore, in the production of safer food products. Two of the most important foodborne pathogens are Staphylococcus aureus and Salmonella enterica ser. Typhimurium, and, hence, the study of their ability to grow in biofilms is of great significance. Alongside, the last years, an increased interest has been placed in the use of beneficial microorganisms, ehxibiting antimicrobial activity, for the purpose of enhancing food safety. Streptococcus macedonicus is a bacterium with high exploitation potential in the field of biopreservation, mainly through bacteriocin production. In this context, the objective of this study was the evaluation of the biofilm-forming abilities of the pathogens Staph. aureus and S. Typhimurium, in the presence of cells or metabolic products of Str. macedonicus. Initially, the biofilm formation behavior of the abovementioned pathogenic bacteria was assessed on stainless steel coupons, in both monospecies and mixed (i.e. dual species, suspensions of each pathogen with Str. macedonicus) bacterial suspensions. Cultures were incubated at 37 and 15°C for a total of 72 and 144 h, respectively. Cell attachment in each type of culture was evaluated at 3 h of incubation at 15°C in saline, while biofilm growth was assessed in broth during incubation at the above temperatures. No significant effect of Str. macedonicus on the biofilm growth of the studied pathogens was observed under the conditions of this study. On the other hand, the growth of Str. macedonicus in mixed biofilms seemed to be favored, especially in the presence of S. Typhimurium, despite the fact that the latter pathogen supressed its initial attachment. During biofilm development on coupons under the aforementioned conditions, coupons-samples were received at 72 h for 37°C and at 144 h for 15°C, for each case, for the measurement of electrical conductivity in a Malthus apparatus. The apparatus measures the electrical conductivity, which changes as long as there is growth and therefore, the concentration of metabolic products in the solution alters, and it gives the time value on which it is stabilized. Unfortunately, the volume of the samples that gave a measurable detection time was low, therefore no conclusions could be drawn. Next, the biofilm-forming ability of the studied pathogens also was evaluated (under single culture conditions) in the presence of cell-free supernatant (CFS) of Str. macedonicus at concentrations 0%, 20% and 40% (v/v), in polystyrene microtiter plates, through the utilization of the crystal violet biofilm formation assay and optical density measurements (580nm). The results were consistent with those on growth on coupons, except for S. Typhimurium at 37°C and in the presence of 40% CFS, for which a higher optical density measurement was recorded, indicating a higher biofilm-forming ability compared to the control (0% CFS). Likewise, the biofilm forming ability of the pathogens Staph. aureus and S. Typhimurium was evaluated in monoculture conditions in the presence of 0% and 40% (v/v) CFS of Str. macedonicus, on stainless steel coupons. Biofilm formation was evaluated during incubation at 37 and 15°C for a total of 72 and 144 h, respectively. The presence of CFS did not seem to affect the biofilm development of Staph. aureus, but it kept the growth of S. Typhimurium at lower levels in the presence of 40% CFS compared to its absence (0% CFS). Furthermore, at 37οC, the observed attachment (3 h of incubation) was better than that observed at at 15°C, and the presence of 40% CFS seemed to affect the attachment of both pathogens compared to the control (0% CFS). The collected data demonstrate that biofilms are dynamic systems, and that various factors may affect considerably their behavior. Hence, further research regarding the effect of different combinations of intrinsic and environmental parameters on biofilm development is warranted, and is expected to contribute to a better understanding of the underlying mechanisms as well as to the development of effective control interventions.
