Ο διεθνής και συνεχόμενος ανταγωνισμός στην αγορά οίνου, σε συνδυασμό με τις απαιτήσεις του καταναλωτικού κοινού για νέα, πολύπλοκα και ενδιαφέροντα προϊόντα, έχουν δημιουργήσει στην επιστήμη της Οινολογίας, νέες προκλήσεις. Η ενασχόληση με αλκοολικές ζυμώσεις χωρίς την παρουσία του μικροοργανισμού Saccharomyces cerevisiae αποτελεί μία υποσχόμενη απάντηση σε αυτές. Επομένως, είναι σημαντική η διερεύνηση των ειδών των μικροοργανισμών που μπορούν να συμμετάσχουν στη διαδικασία παραγωγής οίνου.
Εκκινώντας από την ικανότητα των μικροοργανισμών να καταβολίσουν συγκεκριμένους υδατάνθρακες, στην παρούσα μεταπτυχιακή διατριβή, επελέγησαν οκτώ στελέχη, τα οποία ερευνήθηκαν ως προς την ανάπτυξή τους σε συγκεκριμένη ποσότητα σακχάρου και απολύτως αερόβιες συνθήκες. Εν ολίγοις, πρόκειται για τη μελέτη του φαινομένου Crabtree στα στελέχη αυτά. Μετά από δύο σειρές πειραμάτων στις οποίες ακολουθήθηκαν ακριβώς οι ίδιες διεργασίες, προέκυψαν συμπεράσματα ως προς την ικανότητα των μικροοργανισμών να εκτελέσουν αλκοολική ζύμωσης στις συνθήκες του πειράματος, αλλά και πολλά άλλα ενδιαφέροντα στοιχεία της ανάπτυξής τους.
Οι μικροοργανισμοί που χρησιμοποιήθηκαν είναι οι ακόλουθοι: Saccharomyces cerevisiae, Candida boidinii, Candida oleophila, Candida tropicalis, Metschnikowia pulcherrima και Williopsis saturnus που αποδείχθηκαν θετικοί ως προς το φαινόμενο Crabtree και οι μικροοργανισμοί Yarrowia lipolytica και Pichia ciferrii που αποδείχθηκαν αρνητικοί. Στην πρώτη σειρά πειραμάτων το υπόστρωμα αποτελούνταν από γλυκόζη 60g/L και στη δεύτερη συνυπόστρωμα αποτελούμενο από γλυκόζη 30g/L και φρουκτόζη 30g/L. Οι μικροοργανισμοί αναπτύχθηκαν σε θερμοκρασία 29οC και σταθερό pH = 6.
International and continuous competition in wine market, along with the demands of consumers for new, complex and more interesting products have created new challenges in the science of Enology. Fermentation with non – Saccharomyces yeasts, is a promising answer to these challenges. For this cause, it is important to find the yeast strains that can be used in the wine making procedure.
Starting from the ability of microbes to uptake specific hydrocarbons, in this study, eight different yeast strains were examined under aerated conditions, and the same amount of sugars as substrates. In other words this thesis is an investigation of the Crabtree effect for these yeasts. Two different experiments took place and in each one of them, the same procedures were occurred. The conclusions obtained were about the ability of the yeast strains to carry out alcoholic fermentation in these conditions and also many other interesting facts about their metabolic activity.
The yeast strain used were the following: Saccharomyces cerevisiae, Candida boidinii, Candida oleophila, Candida tropicalis, Metschnikowia pulcherrima and Williopsis saturnus, which proved to be Crabtree positive, and Yarrowia lipolytica and Pichia ciferrii, which proved to be Crabtree negatine. Glucose 60,0 g/L was used as substrate in the first experiment and a blend of Glucose 30,0 g/L and Fructose 30,0 g/L in the second. Fermentations performed in 250 mL flasks, containing 50 mL of the culture medium, T=29 oC and the pH was maintained to the value of ∼ 6,0.
