Ένα από τα σημαντικότερα προβλήματα που αντιμετωπίζει ο αγροδιατροφικός τομέας είναι η μειωμένη αποτελεσματικότητα πολλών Φ.Π. λόγω της ανάπτυξης ανθεκτικότητας φυτοπαθογόνων οργανισμών, εντόμων και ζιζανίων σε αυτά. Ανάμεσα στους μηχανισμούς με τους οποίους οι οργανισμοί αναπτύσσουν ανθεκτικότητα σε Φ.Π., αυτός της αυξημένης απέκκρισης μέσω ABC πρωτεϊνικών μεταφορέων δ.ο. με διαφορετικό μηχανισμό δράσης, παρουσιάζει αυξημένο ενδιαφέρον για την ερευνητική κοινότητα. Οι τελευταίες εξελίξεις στο τομέα των «ομικών» επιτρέπει πλέον την εις βάθος μελέτη του μεταβολισμού ενός οργανισμού και τις επίδρασης που έχουν σε αυτόν καταπονήσεις. Στα πλαίσια αυτά, χρησιμοποιώντας το σακχαρομύκητα Saccharomyces cerevisiae ως οργανισμό-μοντέλο, σε συνδυασμό με την εφαρμογή προηγμένης μεθόδου μεταβολομικής/βιοπληροφορικής, μελετήθηκε η εμπλοκή του ABC μεταφορέα YCF1, ο οποίος βρίσκεται στα κενοτόπιο των κυττάρων, και του PDR5, ο οποίος βρίσκεται στην κυτταρική μεμβράνη, στην ανθεκτικότητά του σε επιλεγμένα μυκητοκτόνα. Τα αποτελέσματα της GC/EI/MS μεταβολομικής έδειξαν ότι η μέθοδος έχει μεγάλες δυνατότητες στην υψηλής ρυθμό-απόδοσης (high throughput) μελέτη του φαινομένου της ανθεκτικότητας μυκήτων σε μυκητοκτόνα. Από τις δ.ο. που μελετήθηκαν, το τριαζολικό flusilazole, διαπιστώθηκε ότι πιθανόν αποτελεί υπόστρωμα του μεταφορέα YCF1. Με την εφαρμογή GC/EI/MS μεταβολομικής καταγράφηκε η επίδραση του γονοτύπου και της καταπόνησης μετά την εφαρμογή του flusilazole στο μεταβολισμό του S. cerevisiae. Διαπιστώθηκαν διακυμάνσεις στη δραστηριότητα βιοσυνθετικών μεταβολικών οδών συσχετιζόμενων με αντιδράσεις σε καταπονήσεις και ανακαλύφθηκαν αντίστοιχοι μεταβολίτες-βιοσημαντές. Ανάμεσα σε αυτούς οι a,a-trehalose, glycerol, myo-inositol-1-phosphate, GABA, L-glutamine, L-tryptophan, L-phenylalanine, L-tyrosine, phosphate, οι οποίοι πλέον του ρόλου τους στις αντιδράσεις οργανισμών σε καταπονήσεις, λειτουργούν και ως σήματα για την ρύθμιση του μεταβολισμού τους. Από όσο γνωρίζουμε δεν υπάρχουν έρευνες σχετικές με το ρόλο του YCF1 μεταφορέα στην ανθεκτικότητα μυκήτων σε Φ.Π. |

Among the major challenges that the agri-food sector is facing, is the reduced efficiency of several Plant Protection Products (PPPs), due to the development of resistant populations of phytopathogens, insects, and weeds. One of the mechanisms that leads to fungicide resistance is the efflux of the active ingredient, which is regulated by ATP-binding cassette transporters (ABC transporters). This mechanism has attracted the interest of researchers during the last two decades. Nonetheless, the latest developments in the so-called "omics" technologies enable the in-depth study of the metabolism of an organism and its fluctuation in response to various stimuli. Within this context, using yeast (Saccharomyces cerevisiae) as model organism, in combination with advanced metabolomics / bioinformatics, the involvement of the ABC transporter YCF1, which is located in the vacuole membrane, and that of the PDR5 transporter, which is imbedded in the cell membrane, in fungicide resistance was studied. The results of GC/EI/MS metabolomics confirmed the robustness of the method and its applicability in the high-throughput study of fungal resistance to fungicides. Amongst the fungicides being studied, flusilazole, which belongs to the azole group of DMIs inhibitors, was discovered as a possible substrate for the YCF1 transporter. Analyses also reviled the effect of genotype and fungicide application on the metabolic profile of S. cerevisiae. Fluctuations in the activity of various biosynthetic metabolic pathways associated with stress responses were recorded and corresponding metabolites-biomarkers were discovered. Among those, the metabolites α, α-trehalose, glycerol, myo-inositol-1-phosphate, GABA, L-glutamine, L-tryptophan, L-phenylalanine, L-tyrosine, and phosphate, were the major ones. Such metabolites play various roles in fungal physiology, and among others some of those serve as signaling molecules in metabolism regulation. To the best of our knowledge, this is the first report on the implication of YCF1 on fungal resistance to PPPs. |
