Τα παραδοσιακά γαλακτοκομικά προϊόντα σκιαγραφούν την παράδοση του κάθε τόπου, στον οποίο παράγονται, καθώς έχουν μοναδικά χαρακτηριστικά. Παράγονται συνήθως με παραδοσιακές μεθόδους και δεν υφίστανται επεξεργασία, όπως συμβαίνει στις βιομηχανίες. Για τους παραπάνω λόγους επιλέχθηκαν 2 τυριά , το Γκερεμέζι (παραδοσιακό τυρί Αίγινας) και το Καρίκι ( παραδοσιακό τυρί Τήνου). Και τα δυο αυτά προϊόντα αποτέλεσαν αντικείμενο της μελέτης με σκοπό την εύρεση οξυγαλακτικών βακτηρίων γνωστών για τις πολύτιμες ουσίες που παράγουν και είναι ιδιαίτερα ευεργετικές για τον ανθρώπινο οργανισμό. Το διαφορετικό στοιχείο που αποτέλεσε ιδιαίτερο ενδιαφέρον παρούσα μελέτη ήταν το γεγονός ότι όλες οι καλλιέργειες που μελετήθηκαν προήλθαν από την παρασκευή των τυριών, χωρίς προσθήκη εμπορικών καλλιεργειών. Στην παρούσα μελέτη ερευνήθηκε το μικροβίωμα των παραπάνω τυριών. Τα απομονωθέντα στελέχη βακτηρίων και ζυμών αρχικά ομαδοποιήθηκαν με τη μέθοδο rep-PCR και στη συνέχεια, αντιπροσωπευτικά στελέχη βακτηρίων και ζυμών ταυτοποιήθηκαν με την αλληλούχηση του γονιδίου 16S rRNA και της περιοχής ITS DNA, αντίστοιχα. Ταυτοποιήθηκαν κυρίως είδη των γενών Staphylococcus, Streptococcus, Lactobacillus και Pichia.
Traditional dairy products always attribute special features to the place where these are produced due to their unique characteristics, the traditional methods with which these are produced and the fact that these don’t undergo processing as it happens in industries. The above reasons set off the study of 2 traditional cheeses, Geremezi (product of Aigina) kai Kariki (product of Tinos). Both products have been the subject of study with purpose of finding LAB which produce beneficial substances for human such as lantibiotics (widely known substances with antimicrobial role). The special element of these two products is the fact of no added starter cultures. In the current study it was investigated the ‘’microbiota’’ of the above products. PCR techniques were used to group and then identify the isolated microorganisms. The isolated bacteria and yeasts were grouped using the genotyping technique of rep-PCR. Then, representative strains of bacteria and yeasts were identified at the species level by sequencing the 16S rRNA gene and ITS DNA region, respectively. The results obtained from the sequencing of 16S rRNA gene and ITS DNA region were analyzed using specific tools of bioinformatics and showed mostly strains of Staphylococcus, Streptococcus, Lactobacillus and Pichia, concerning yeasts.
