Σκοπός της παρούσας μελέτης ήταν η διερεύνηση της βιοχημικής δυνατότητας των στελεχών Cunninghamella echinulata και Mortierella isabellina, ανηκόντων στην τάξη Mucorales, να συσσωρεύσουν μικροβιακό λίπος με σημαντικές ποσότητες γ-λινολενικού οξέος (GLA, A6912C18:3), ένα πολυακόρεστο λιπαρό οξύ υψηλής φαρμακευτικής και διατροφικής σημασίας, κατά την καλλιέργειά τους σε σακχαρούχες ανανεώσιμες πηγές άνθρακα. Τα υποστρώματα που χρησιμοποιήθηκαν ήταν η εμπορική γλυκόζη, η εμπορική φρουκτόζη, η εμπορική ζάχαρη και η μελάσα, σε αρχική συγκέντρωση 60 g/L. Όλες οι ζυμώσεις πραγματοποιήθηκαν σε καλλιέργειες κλειστού τύπου, ενώ ο λόγος C/N ήταν σε κάθε περίπτωση υψηλός (200 moles/moles), ώστε να κατευθυνθεί ο μικροβιακός μεταβολισμός προς την σύνθεση αποθηκευτικού λίπους. Κατά την διεξαγωγή των πειραμάτων στην εμπορική γλυκόζη, και οι δύο μύκητες παρουσίασαν ικανοποιητική ανάπτυξη και παραγωγή λίπους με σημαντικές ποσότητες GLA. Ειδικότερα, ο μύκητας Cunninghamella echinulata παρήγαγε 12.9 g/L βιομάζα, με 3.8 g/L λίπος και 740 mg/L GLA, ενώ ο Mortierella isabellina 13.2 g/L βιομάζα με 9.85 g/L λίπος και 345 mg/L GLA. Στην εμπορική φρουκτόζη, ο μεν Cunninghamella echinulata παρήγαγε 16.7 g/L βιομάζα με 3.6 g/L λίπος και 360 mg/L GLA, ο δε Mortierella isabellina παρήγαγε 11.4 g/L βιομάζας, με 7.36 g/L λίπος και 110 mg/L GLA. Ωστόσο, ενώ στην εμπορική ζάχαρη ο Cunninghamella echinulata αναπτύχθηκε ικανοποιητικά (16.6 g/L βιομάζα, 3.1 g/L λίπος και 435 mg/L GLA), o Mortierella isabellina απέτυχε να αναπτυχθεί επαρκώς (5 g/L βιομάζα και 0.5 g/L λίπος). Στο εν λόγω υπόστρωμα μετρήθηκε και η ενεργότητα της εξωκυτταρικής ιμβερτάσης, με μέγιστη τιμή 0.5 U/mL από τον Cunninghamella echinulata. Επιπλέον, στο υπόστρωμα της μελάσας και οι δύο μικροοργανισμοί αναπτύχθηκαν επαρκώς (12.1 g/L βιομάζας με 3.8 g/L λίπος και 420 mg/L για τον Cunninghamella echinulata και 10.1 g/L βιομάζα με 4.8 g/L λίπος και 168 mg/L GLA για τον Mortierella isabellina). Στο εν λόγω υπόστρωμα παρατηρήθηκε έντονος αποχρωματισμός της μελάσας, ήτοι 75%, από τον μύκητα Cunninghamella echinulata. Τέλος, πραγματοποιήθηκε περαιτέρω επεξεργασία των λιπιδίων του μύκητα Mortierella isabellina, κατά την κλασμάτωση και ανάλυση τους στον αέριο χρωματογράφο. Παρατηρήθηκε ότι το κλάσμα των ουδέτερων λιπιδίων παρουσίαζε ομοιότητες ως προς την σύσταση του σε σχέση με το ολικό λίπος, ενώ το κλάσμα των φωσφολιπιδίων περιείχε μεγαλύτερες ποσότητες ακόρεστων λιπαρών οξέων (και κυρίως γ-λινολενικού οξέος), σε σχέση με το ολικό λίπος του Mortierella isabellina.
Aim of the presen study was to investigate the biochemical potentialities of two
mould strains belonging to the order of Mucorales, in particular Cunninghamella
echinulata and Mortierella isabellina, of accumulating single cell oil containing
important quantities of -linolenic acid (GLA,
C18:3), an unsaturated fatty acid
of significant medical and nutritional importance, during their growth on sugarbased
renewable
carbon sources.
The substrates
used
for
this
purpose were
industrial
glucose,
industrial
fructose,
industrial
sugar and molasse, in
an initial
carbohydrates
concentration
of
60
g/L.
All
cultures
were conducted
in
batch-flask
type,
while
in
all
experiments,
nitrogen-limited
media
were
employed
in
order
to direct
the microbial
metabolism
towards the synthesis
of reserve
lipid (initial molar C/N
ratio 200
moles/moles).
In
the case of industrial
glucose,
both strains
grew satisfactory.
Specifically,
Cunninghamella echinlata produced 12.9 g/L biomass, with 3.8 g/L of
lipid, containing 740 mg/L GLA, whereas Mortierella isabellina produced 13.2 g/L
biomass with 9.85 g/L of lipid, containing 345 mg/L. Growth on commercial fructose
resulted in 16.7 g/L of biomass with 3.6 g/L of oil, containing 473 mg/L GLA for
Cunninghamella echinlata, wheras Mortierella isabellina produced 12.1 g/L biomass,
with 7.36 g/L of oil containing 110 mg/L -linolenic acid. Mortierella isabellina failed
to grow satisfactory on industrial sugar [biomass produced of 5.0 g/L containing
10% (wt/wt) lipid] with negligible quantities of extra-cellular invertase produced,
while Cunninghamella echinlata yielded 17.6 g/L of biomass, with 3.1 g/L of oil
containing 310 mg/L. Also, Cunninghamella echinlata produced a maximum of 0.5
U/mL of extracellular invertase. Moreover, both moulds showed remarkable growth
on molasse [12.1 g/L of biomass with 3.8 g/L of oil containing 490 mg/L GLA for
Cunninghamella echinlata and 11.3 g/L of biomass with 4.8 g/L of oil containing 168
mg/L GLA). Worth noticing is the fact that Cunninghamella echinlata decolozised the
medium containing molasse up to 75%. Last but not least, further analysis via
fractionalization was conducted in single cell oil obained from lipid-accumulating
period of Mortierella isabellina, showed that the composition of fatty acids of the
neutral fraction resembles with that of total lipids. Phospholipids contained a somehow higher quantity of unsaturated fatty acids (and principally -linolenic acid)
compared with total Mortierella isabellina lipid.
