dc.description.abstract | Το επιστημονικό πεδίο που μελετά τις επιπτώσεις της ηλεκτρομαγνητικής ενέργειας έχει αναπτυχθεί σχετικά πρόσφατα και συνεχίζει να αναπτύσσεται ραγδαία σε παγκόσμια κλίμακα. Καθώς η σύγχρονη χρήση ηλεκτρομαγνητισμού έχει λάβει ανεξέλεγκτες διαστάσεις κρίνεται απαραίτητο η βαθύτερη γνώση αυτού του φαινομένου. Οι μέχρι τώρα επιστημονικές αναφορές έχουν χαρακτηριστεί με έλλειψη συνέπειας στα ευρήματα και αδυναμία συσχέτισης των διαθέσιμων αποτελεσμάτων. Παρά τις εκτεταμένες προσπάθειες για σαφή συμπεράσματα, έως σήμερα δεν έχουν επικρατήσει ξεκάθαροι μηχανισμοί αλληλεπίδρασης ηλεκτρομαγνητικού πεδίου και κυτταρικών οργανιδίων. Στην εισαγωγική ενότητα της παρούσας εργασίας επιχειρείται μια συνοπτική αναφορά των βασικότερων απόψεων που έχουν επικρατήσει πάνω στη δράση ηλεκτρομαγνητισμού in vitro με βιολογικά στοιχεία. Επίσης γίνεται μια σύντομη αναφορά των κυριότερων εννοιών της ηλεκτρομαγνητικής ενέργειας και του ηλεκτρομαγνητικού πεδίου, καθώς και των κυριότερων μεθόδων παραγωγής ηλεκτρομαγνητισμού in vitro. Βασικός στόχος της πειραματικής διαδικασίας είναι η εξαγωγή τυχόν συμπερασμάτων μέσω μιας προκαταρκτικής μελέτης, με κύτταρα νευροβλαστώματος N2a και ινοβλαστώματος Vero υπό ακτινοβολία παλμικής ηλεκτρομαγνητικής ενέργειας υψηλών συχνοτήτων (27,12 MHz). Το παρόν πεδίο επάγεται από μια πειραματική συσκευή σε σχήμα δακτυλίου κατασκευασμένη για κλινικές εφαρμογές από το πανεπιστήμιο Πατρών. Περιγράφονται αναλυτικά στην ενότητα των μεθόδων όλες οι διαδικασίες χειρισμού των κυτταρικών σειρών που ακολουθήθηκαν από την απόψυξη, την καλλιέργεια και τον υπολογισμό του αριθμού τους, καθώς και οι τρεις μέθοδοι, η χρωματομετρική μέθοδος ΜΤΤ, η μέθοδος trypan blue για την καταμέτρηση των νεκρών κυττάρων και η μέθοδος τραύματος δι' αμυχής (scratch - wound assay) για τη μελέτη της μεταναστευτικής ικανότητας. Στο πειραματικό μέρος περιγράφονται αναλυτικά όλα τα βήματα και οι χρόνοι εκτέλεσης των πειραματικών προσπαθειών. Τέλος, βάσει των αποτελεσμάτων δεν διεξήχθησαν ασφαλή συμπεράσματα για τη δράση του παρόντος υψίσυχνου ηλεκτρομαγνητικού πεδίου στις συγκεκριμένες κυτταρικές σειρές. | el |
dc.description.abstract | The development of the scientific field that studies the consequences of the electromagnetic energy is recent and it is growing rapidly on a global scale. The deeper knowledge of electromagnetism is needed, because of the highly increased usage of it in our days. Because of the deviance in the findings of the scientific reports, correlation between results is not feasible. Despite the extended efforts for outright results, till today there is no clear type of interaction between electromagnetic field and cellular organelles. At the opening chapter of this particular research there is a summary report on the basic and dominant aspects regarding the in vitro effect of the electromagnetism on biological elements. Also, there is a short report on the main concepts of electromagnetic energy and electromagnetic field, as well as the main production methods of electromagnetism in vitro. Primary aim of the experimental procedure is the conclusion through a preliminary study of N2a neuroblastoma cells and Vero fibroblast cells under high frequency pulsed radiation of electromagnetic energy (27.12MHz). This field is generated from a ring shaped experimental device that is built by the University of Patra for clinical applications. In the chapter about methods there is a precise description of the management procedures of the cell lines during their defrost, their culture and the calculation of their number, along with three methods, the colorimetric method MMT, the trypan blue method for the measurement of dead cells and the trauma by hack method (scratch - wound assay) for the study of the migration ability. In the experimental part there are all the steps that have been used and the run time of all experimental efforts. Finally, according to the results there were not safe conclusions about the effect of the high frequency electromagnetic field on the particular cell lines.
